Can anyone guide me about method to measure the Activity of

Carbonic Anhydrase (EC4.2.1.1 CA) enzyme in plant leaves by spectrophotometer ?

Do you want to measure the activity of the enzyme while it is still in the leaves, or are you going to make an extract of the leaves?

There is literature on measurement of CA activity in leaf extracts that should provide guidance. Here are two examples.

You also have to think about which part of the leaf you are investigating, and which subcellular compartment.
