I am working in cerebral ischemic models. I generally used in vivo models in mice or rat for the ischemia induction. I am also interested in in vitro ischemic models such as OGD-R. Would be very grateful if anybody could provide me the detail of the ogd protocol.
Hi Bhakta...you can get the solutions you need to do OGD in primary neuronal cultures on our web site (link below)
It includes how to make cultures, feed them and check them for viability as well as the reagents needed to expose them to OGD.
Once you have cultures established, you can use a Billups Rothenberg hypoxia chamber hooked up to a nitrogen tank and expose cultures for as long as you like. In mature neuron enriched cultures , you can see cell death 24h after a 30-60 minute exposure.
Neuronal culture is quite an art form, so it will be time intensive to set up. Good luck.
