Hi Tushar, what concentrations were you using in your exposures and what life stage of the zebrafish did you test? I have worked with the effects on anti-androgens and oestrogens in the gonads of medaka so I would be interested to have a look at your histology if you think I can assist. I have also observed fibrosis as Victor has mentioned above so this might be something to look for.
Can you do the study in mice for regulatory purposes?
To test the reproductive toxicity of a compound as plasticizers, first determine the specific compound, determine the optimum dose that you expect to produce a phenotype, the route of administration to zebrafish, the duration of drug administration, are you going to study the effect on gonad development? this includes spermatogenesis, testis, male reproductive glands, ovary, oocyte and uterus development. You can also study the effect on reproductive functions and fertility. You can do a microarray or proteomics to determine the effect on target gene expression. Important genes include but are not limited to the study of ER, PR, AR. The HPG axis,hormonal levels are also interesting to study. You can study the effect on sexual development, feminization or masculinization of males or females.
It is very complicated and impractical to study all these aspects in one project, better to focus on one sex as male or female zebrafish, then determine if there is an effect of the chemical and examine the cellular and molecular mechanism of toxicity.
