I have wound samples from porcine skin, fixed in PFA 4% and embedded in paraffin, is there any particular H&E stain that is more suitable for this? Many thanks
Thank you very much for your answers, no I haven't tried any yet, I was just wondering was any in particular more suitable. I think we have Mayers in the lab so will give that ago. Many thanks
All types of hematoxlin are OK, just filter it before use. immerse the slides for 15 min. in hematox. the wash for 10 min n tap water. then put them in eosin for 10 min. and wash for 30 second.
Try fixing your sample in 10% neutral buffered formalin. 4% paraformaldehyde is not the best fixative for H and E. If you need the milder fixation for other procedures, take two samples.
I would suggest that you contact a histology lab and have them do the staining. I'm sure you can get some staining done by yourself, but a proper histology lab will have the protocols worked out and the standardization done so that the staining is optimal for evaluation. I have seen too many times suboptimal stains done by neophytes that cannot be properly evaluated because of poor contrast or other issues.
Also if you are planning on evaluating fibrosis in your wound sections, a basic stain for collagen would be appropriate, like a Masson trichrome. Again, a basic histology lab would have the technique worked out.
