We are doing research work by using fungus. What is the problem raising here means on the molecular identification ITS regions notes one of the species and 18rRNA typing gives another one and we are also trying laboratory growth and we observed some extent of growth. I've tagged photos here, can anyone help in this regards?
Thank you sir for reply to my question we doing both ITS and 18sRNS sequence one gives microporus affinis and another one notes agaricomycetes but phenotypically massive non sporulated white color fungus but it gives a morphologically a bow type structure
Maybe you could not get the pure isolate.
The colonial characters of the isolated strains might show the information about this.
If you have the pure culture in media, can you do DNA isolation and sequencing again?

You have whole complete 18sSSU and ITS sequences, which identified, with BLAST, to get different specie result, right? Can you check the similarity of these sequences compare to those species? If they are still 100% similar to those species, maybe you have to do D1D2 of LSU sequencing, primers LROR & LR7(or LR5), to confirm the identification.
sorry sir we are doing D1 and D2 domines and 18s rRNA sequence they give different species and grown for fruiting development in our lab but we doesn't get and show some extent of growth as shown a bow

I am agree with Dr. Jiri Kout, your micelia belong a Polypore fungi, because it form a basidiome with pores in your pictures. And one region determinate this as Microporus affinis and this is a Polyporoid fungi.
Did you determinated this micelium with ITS as Microporus affinis, but not with 18S?, 18s is not a good region in poliporoid fungi to determinate species, It is generally used for Higer Taxa Phylogeny. You have to use others regions as 28S, tef, or RPB2 that are better regions to determinate polyporoid fungi.
Where did you get this mycelia? This is important to determinate this fungus, too.
I think, If you observed some structures in Mycroscope, doing preparations in the poroid part of your culture, may be, you can see some structures (spores, basidia, cystidia, micelia with or without clamps conections, skeletal hyphae, etc. It is possible to determinate your polyporoid fungi.

In fact, 18rRNA is not suitable to identify a polypores. There is not enough 18rRNA sequences in GenBank. LSU is suitable to do analysis for high level phylogeny. ITS is better than 18rRNA and LSU, and you can trust the result of BLAST from ITS sequence. I suggest you do slice observe to determine that which is the species in Microporus.
