I have ordered Chlamydomonas reinhardtii strain from ATCC Company and in next days it will arrive for me. in fact I have no any experience for culture microalgae and this time wil be for first time. I have read too much paper to find different dimension of culture condition of them and how we can maintenance them. But there is a problem that I couldn't find it's answer. I want to know that when This strain arrive for me, after thawing the sample, how much I should take strain and input in my TAP liquid medium?. I want to culture it in TAP medium.
please help me if any one has same experience.

Anyway ....dont waste your culture suspension , try to start a culture adaptation by 10% of concentration ! if your succeed to maitain your strain after few days ,you can fix the conditions you need to !

Chlamy loves TAP so you no need to worry at all. I used to order strains from Chlamy culture, they were quite active as they grew straight on TAP/light. I got the way to prepare TAP stock which quite easy and save time. Let me know if youwould like to have it.

Be careful during work beacuse contamination can occur in TAP medium easily. Best regards.
If you need fast growing and generous morphology , you can use 3NBBM-V media , boosted by Thiaminhydrochloride and Cyanocobalamin (vitamins B2-B12) , I promise you can visualize a spectacular circular movements from your microscopic ocular ....but I prefer axenic culture as the media is powered by vitamins (!!!)
