I am doing phenol-chloroform DNA extraction of FFPE tissue. We have a standardized protocol we use regularly. I have my tissue samples in lysis at the moment, TES buffer + Prot K. My question is, that if unable to continue due to unforseen events, can these lysates be saved and stored at say -20C and continue later? Samples are unique and most likely will not be able to aquire more to redo extraction.
If the tissues are hard to get then don't even think about it, prioritize them and finish the extraction. Pure DNA is far more stable than crude lysate. Storing lysate at -20 for more than a day degrades the quality and yield of DNA. I have had the experience where I was unable to PCR amplify a section of DNA from lysate stored for 2days, while I was successful with DNA from fresh lysate.
