when I perform enzyme kinetics experiments with my enzyme, the data fits with hills equation (sigmoidal curve with R square value of 99% ). However my enzyme is monomeric and it has single substrate binding site. Can it be possible?? If so can anyone give a reference for it preferably an enzyme which works on a polysaccharide.

Sigmoidal curves are usually observed for cooperative behavior of quaternary proteins. For these proteins, n > 1 from Hill's plot. If n = 1, there is no cooperativity. What is the value of your Hill's constant?

The answer is yes. There are more tan one model to explain this behaviour. For example that proposed by Ricard, the nemonic enzyme.

I read this article and it tells about conformation change in the enzyme for sigmoidal kinetics....
