In single molecule fluorescence imaging, the dye molecules are irreversibly damaged by the excitation of photons, and the molecule fluorescence loses. If there is a molecule that has absorption peak, but with little or no fluorescence. Under laser irradiation (in the range of absorption wavelength), do the molecule be damaged or "photobleached"?

Yes. Emission and photo-bleach are different de-activation pathway, like you can either walk or ride to your office.
The mechanism of photo-bleaching can be complicated. In many cases, the observation will indicate a first order photobleaching, and singlet oxygen is believed to play important roles in such first order photo-bleaching. However, to the best of my knowledge, no comprehensive mechanism has been put forward for higher order photo-bleaching.
