Hi researches out there. Does anybody know if you can read a glucose colorimetric assay using hexokinase reagent at wavelengths >400 nm? Please let me know if you have done that and what wavelength worked for you. My plate reader can only do 405 or higher.
John F. Odhiambo This completely depends on the substrate. So we'd need to know a little bit more about the assay to give you concrete suggestions.
However, if you don't have the optimal filter (combination) available to measure at the optimal wavelength, a wavelength close to it usually works fine, just the sensitivity will be lower. Recording a UV/Vis spectrum of the assay reagent before and after development will give you a lot of information. In worst case, run the reactions in cuvettes and use a spectrophotometer.
