hi,
today I found out that some aliquots of primary antibody has been dried and only a sign of it is visible on the bottom of the vials.its looks like a pellet. can i use this antibody or I should throw it away?
I have storaged them in 4 degree refrigerator.
I appreciate your wise and kind advises.
I would reconstitute the dried "pellet" with just water (not buffer) since water is the only thing that was lost, the original salts should still be there.

I think if you add a few drops of diluent to the vial and mix, then try to do your experiment. Might be it work.
As noted by Hassani, I would add 10 microliter (or the original volume if known) of sterile PBS, vortex the tube and incubate on ice for 30 min. Centrifuge the tube and use the antibody solution for any experiment.

Probably meanwhile solutions to the problem were found. I just want to add that I agree with the proposal of Bruno because his argument seems logic to me.
Once, it happened to me, I just added some microliter of aqua distillate. When you are going to use it depend on your usage amount already, use twice, check the quality if it works!! For me worked well.

I've had multiple experiences with unintentionally [and intentionally] lyophilized antibody. If you know what the original volume of the sample was, you can add that amount of water - I typically warm it to 37oC [remember, that's where these proteins normally are at] for ~30 minutes, microfuge the tube [10-12,000 rpm for ~5 minutes] to remove any ppt, and transfer to a new tube. When I was intentionally lyophilizing purified antibody, the effect on titer [neutralization of colony forming activity] and FACS staining were within a factor of 2 of the original values.
As others suggested you should try. It happens once to me as well and I reconstituted in water and did western blot but the antibody did not work. Wish you good luck.
