Hi, I was running my Western blot with cell lysates and exosome samples and exposed the blot to Calnexin primary antibody (1:500 dilution). As you may know, Calnexin (67 kDa molecular weight) is a negative marker of exosome and positive for cells. Which means I should be able to see bands only for my cell lysates, not exosome samples. However, when I imaged it, the only bands that were present in cell lysates and not in exo samples were around 37-50 kDa. I’m quite confused as Calnexin bands should be around 70-95 kDa. Can anybody please help me understand what went wrong? I have included the blot image here - starting from extreme right is the marker, followed by cell lysate sample and 5 exosome samples. It’s not the best image but you’ll get the idea. Thank you in advance
