Callus induction from the eyes or from the tip of the sprouts will be easier than as compared to induction from the tubers. Changes in the hormone concentrations (in general, lower concentration of auxin and gibberellic acid) may result in direct organogenesis without callus formation.
i am a scholarship student i have limited time. therefore i want to induce callus from sprouts. then i will start adaptation process for tolerance in callus by giving different regimes of salts and Osmolytes by successive subculturing.
You can prepare a semi-solid media, such as MS with thidiazuron immersed in cotton. To the semi-solid media, add some nodes and internodes, around 4-5 for each explant. Grow this in dark. After four-five weeks you can get microtubers. Make thin slices (1 mm thickness) of each microtuber and have them as explants for your callus induction. Your calli should be ready in about 3-4 weeks. Chances are that you may get shoots from eyes, which can grow in 2-3 days. For transgenic generation, you can cut them away.
