Convection-enhanced delivery (CED) system is a powerful treatment for directly bypass BBB in brain tumor therapy. I try to set up the system and the in vivo animal model. In the begin, we use TMZ ( Temozolomide) to test the IC50 value of U87 cells. the IC50 of U87 cells is 250 uM. however, I have no idea...how to decide/calculate the concentration of TMZ ( I think the concentration should be higher than 1000uM...) for CED injection (10ul, 0.5ul/min,directly inject drugs into brain tumor, not IV or IP injection, nude mice, xenograft model)?

is anyone have similar experience or suggestions? I really hope I can have a right direction to do the animals experiments without scarifying unnecessary "testing animals...."
