My lab is experiencing a new issue with several mouse brain tissue sections (for confirming placement, no immunohistochemistry). Each tissue section has white "sparkles" that dissipate over time after mounting using a no-dapi mounting media. We suspect that it may be the tissue, as older perfused brains do not have this issue when sectioned, and that it may take a while for the tissue to absorb the mounting media? We also tried placing the tissue in cryo preservative and 1x PBS buffer but they yielded about the same result. My PI is currently having me rinse the sections 3 times in PBS and then soaking them for 5 days before mounting them. Has anyone experienced anything similar and if so how did you fix the issue?
