Hello, I have in culture some BMMC from mice between 6 and 9 weeks. They still have more than 95% viability and purity. They don't react to IgE-mediated anaphylaxis and ciprofloxacin anaphylaxis (when in mice they do). But with C48/80 they degranulate. I have MC with and without the KitD814V mutation. I tried to increase incubation times, concentration of IgE and the allergen (DNP-HSA) and still nothing. Do you know why they don't degranulate?

Another question: I want to study these BMMC metabolism with the Seahorse technique but they don't seem sensitive. I use 1µM for the concentration, also for the glucose there is not a big pic.
