Hello! I have a anti-mouse MHC Class I antibody (anti-H2Kb-H-2Db) that we are trying to prove works in blocking MHC I mediated interactions. We are using SIINFEKL and OTI mice as a model to do so.

I harvest splenocytes from OTI mice, then make into a single cell suspension and incubate for 24 hours with .00001 ug/mL SIINFEKL. This activates around 10% of cells, but no changes with the blocking antibody. Does anybody have any idea why this isn't working? We really need this antibody to work.
