Hello everyone! Anybody worked with HCC-70 cell line before?
I have a "black-dots contamination", and I had read a lot of similar questions, but I still need to ask one (just to be sure about my conclusions).
I tried to incubate conditioned media for 2 days, but no new black dots appeared, while their density in flask with cells increased. So, my conclusion is, that those black particles are exosomes. But I can't find any references in publications about such behaviour of HCC-70. Any suggestions about origin of these dots?
P.S. If anybody interested about media - RPMI-1640, 2mM L-glutamine + 10% FBS
I have the same problem with HTR8-SVNeo...
Media: RPMI-1640, 2mM L-glutamine + 15% FBS (additional HEPES).
Cell growth is not affected.

I am not sure what is happening to your cells, but I don't think the black dots are exosomes. Exosomes are only 100nm in diameter, you need an electron microscope to see them properly, so you really couldn't see them in a normal cell culture microscope. That said, they could be debris from the cells, or even apoptotic bodies. Have you tried adding antibiotics to your media? Erradication or decrease of the black dots could show that. Do you perform mycoplasma screening? It would be helpful if you could upload of photo of what you are seeing in your cells.

HCC70 secrete allot, I don't know if the black dots are exosomes, but they are a natural product of the cell line.
I tested their medium by growing it seperately for a week or so, and saw that nothing grows there when the cells are gone. At some point we purchased fresh cells from ATCC, and they behaved the same.
Thank you, Lee! Right now I looked at incubated media, and yes - there is no black dots in absence of HCC70. So I was right - this is some kind of cell's product.
Hello, Yucheng! No, I didn't solved this problem yet, and found another line from another stock which suitable for my project.
