I'm looking for highly accurate and precise methods of quantifying the amount of phospholipid in either an aqueous or organic mixture. I'm currently using the ammonium ferrothiocyanite assay but I'm looking to more to something that's a bit more consistent and has fewer assumptions as part of its quantification procedure. Any suggestions?

Thank you very much for posting an interesting technical question. Phospholipids (PL) can be quantified in many ways, for example, assaying P, using chromatographic techniques like GC, TLC and HPLC, and using spectroscopic methods e. g. 31P NMR, HPLC-MS etc. However, spectroscopic method especially 31P NMR can be applied to fast and accurate determination of the PL. For detail procedure, please have a look at the following article which has already been posted in RG. If you just want the total concentration of phosphor you can use ICP-OES.
