I conserved the extracted microbial DNA from kit in -20°C (in TE BUFFER), after that i was obliged to transfer the microbial DNA to + 4°C because i can't ensure the transport from a place to another at the same temperature of freeze (-20°C), Now it's at +4 °C, i don't know if i can put it again at -20°C after 5 days at +4°C because i worry about the actions of freeze/thaw that may degrade the DNA. Can i put it again at -20°C knowing that i will use it lately after 1 week ? I need your help as soon as possible thanks
It depends on what you are going to use it for! But if there is any degradation at all, it will be negligible (most of any fragmentation took place during the extraction process). People tend to think that its a delicate molecule that will fall to pieces easily. Its not the case - even if you use a high powered probe sonicator, you still have fragments in excess of 400bp.

Then I don't think you need to worry, unless you are trying to amplify an enormous stretch of DNA! Just make sure you vortex the DNA after thawing - it concentrates at the bottom of the tubes.
