Is it possible that loss of function single gene mutation which do not have any profound effect upon any biological or cellular process on its own, however, show strong effect in the presence of another loss of function single gene mutation? Both genes are components of same pathway.

even if change is very low for gene function due to the presence of a variant, accumulation of gene defaults in the same pathway can cause major results.
try some bioinformatics tools (ingenuity, DAVID, BioGPS...) to see if your target genes are in the same pathways, and check, in regards to the frequencies of the variants in the same sample, to compute the consequence.

Yes, this is very common in organisms with large gene families. You will get functional redundancy and overlap in gene function. Also, sometimes an abnormal phenotype is only apparent in a single mutant during stress (pathogens, cold, etc.).

Dear Dr Katie and professor Khalid, Actually the genetics i found is not so simple. I understand answers from both of you. However, Katie's suggestion about observation of mutant phenotype in a stress condition is practical which i have already tested in some mutants. But In case of my research project, even this is not working. The over-expression and single mutant even T-DNA lines are not showing any significant phenotype but in combination with another single mutant it has very strong phenotype at both cellular and plant growth level.
