Procedure for karyotyping

Karim Baziz

- Roots: from 5 mm upwards in length.

- Pre-treatment: 0.2% colchicines for 1h30- 1h40, at room temperature at 08h-08h30 AM.

- Ethylic alcohol / Acetic acid (3:1) mixture, at room temperature for 24h.

- Hydrolysis: HCl 1N for 4-6 min, at 62°
