In a culture cell system, I have some unexpected result with some cells after drug treatment (induce double strand break). I want to keep these cells in culture, but I want to be sure that the effect is the consequence of the molecule, so that the drug has reached these cells.
I'm looking for a report system which could show me which living cells have received the drug treatment. For example a system where a double strand break lead to the expression of GFP in the living cell.

Some assays allow to evaluate the cell citotoxicity induce by a drug. For example, in de MTT assay, after 24 h of drug exposition the culture cell system is evaluated. Because of, the remarked effects are connect directly with the drug.
Is important also the strict control of the culture cell system variables, in manner that the evaluation are objective.
In my experience, I use one or more normal cell line, non malignant...and others malignant cell line of differents origins. They conform the evaluation system. I observed the double strand break in those that we find apoptosis features.

The MTT lead to the lost of the culture and I want to keep the cells in culture after know if they received the treatment. Others classical DNA break analysis (COMET, TUNEL, gH2AX immunofluorescence), lead also to the lost of the culture.
So I'm looking for other option.
