0.03% Streptomycin is widely used to suppress bacterial growth, when isolating fungi. Unfortunately, in my case, some freaky bacillus spp. keep growing on my Czapek medium, which was meant to select Fusarium spp. from potato.
Hi, I agree with Franci Ago Celar, Streptomycin + penicillin is a good mix. The concentration should be 0.3-0.5 g/L medium for each one. Be sure to add the antibiotic mixture to the autoclaved medium when its temperature is around 50°C and to sterilize the antibiotic mixture by filtration.
Thanks everybody. I added 0.1% lactic acid together with 500mg/L chloramphenicol and it seems to work perfectly. Fusarium mycelium have appeared very well. Waiting further for exudates.
The lactic acid acidifies the culture medium to pH 3.0 and bacteria do not develop in this condition. The results have shown that it is more efficient than adding antibiotic to the culture medium. However, no more than 0.8 ml/liter of medium should be added, otherwise the medium will not solidify.
We are using chloramphenicol (500 mg /L) in all the mycological media, such as Pal sunflower seed medium, APRM agar, Sabouraud dextrose agar and others for the isolation of fungi from clinical as well as environmental samples.The chloramphenicol must be added in the medium after autoclaving when temperature is around 50 degree centigrade. Please check the pH of medium as it should be acidic.If you wish, you can increase the concentration of chloramphenicol by 750 mg per liter.
