You could use a piece of paraffin. Just need a sharp edge in order not to be so dirty and use it like the pap pen. Best of luck!

Are they cells in suspension cultured? If they do, we do the staining with an intracellular flow cytometry protocol and finallywe  put the cells on the slide in aprox. 10 uL of permeabilization buffer. Then we spread the cell suspension, let the cells to dry and add the mounting medium and the cover glass. 

So the dried drop defines the cell area.

PAP pen from sigma is better. I am using this.

Now I'm using Super Pap Pen from Invitrogen and It works very well, althouhg I have to do again the hidrophobic line after PFA fixation. The worst thing is the cost, very expensive.
