Anyone who has worked with culturing of lymphocytes, can u plz tell me that after I isolate the lymphocytes and put them in a CO2 incubator, I need to see if my lymphocytes have grown and whether they are live or not. please help me, that how can I analyse that whether they are Live or death by viewing them under a inverted/light microscope? I mean is there something in the morphology that might indicate whether they are live or have died.

If you just need to observe the cells under the microscope and distinguish the live from dead cells, then the only difference between the two is that live cells are refractile while the dead cells are usually not refractile.
When light hits the cells/cellular parts, some of the light is reflected back and some of the light is absorbed by the cell/cellular parts. In the microscope, what is actually seen is the reflected light. So, in live cells one is able to see the cell morphology & its differences because it refracts (absorbs) some light and reflects the rest. The dead cells do not show this property.

Malcolm Nobre thanks for answering but do u have any pictures or videos where i can see how exactly do live/dead lymphocytes appear under an inverted microscope because i couldn't find any specifically for lymphocytes on the web.
