I am trying to do reaction in non aqueous solvents; i searching for organic solvent to dissolve chemically modified nucleotide triphosphates.
The best way is to form tributyl or tetrabutyl ammonium salt of your triphosphate, then dissolve it in DMF.
If you can not, you can try DMAC (dimethyl acetamide)
But I like the first case.
thanks Ahmed, DMAC is not working but I hope the TBA form of the dNTPs will probably work.
If you can get dNTPs to dissolve in organic solvent by the suggested method, do not expect to be able to achieve as high a concentration in organic solvent as you would make in water.
You can get around mM(or maybe 10mM) for TBA form of the dNTPs in DMF !?
You can make the TBA salt. You just need Dowex-8H+. It is tedious but once you have the salt. you can achieve high concentrations in DMF.
