Some mutation on NGS panels but struggle to determine driver mutation or SNPs.
You can start looking at the COSMIC dataset, but in general this is a difficult task to assess without experimental work. SNVs in the 1000 Genome Project are also expected to be neutral, although this expectation may not always be valid.
try to check all the mutation predictors like CHASM, FATHMM, Mutation Taster, Mutation Assessor and Pholyphen2.
You will have an idea about the mutation if can be potentially pathogenic or not.

Prediction tools for pathogenicity are known to results in lots of false positive and false negative findings.
I suppose finding the driver mutation is totaly not trivial and there will probably not be a standard guideline. You may want to look for example for loss of function in TSG or gene-fusions...
