My model organism is Nostoc punctiforme ATCC 29133. Now a days when ever i am inoculating in 100ml BG11o media pH 7.5 to 8.0 for 1ug/ml Chl-a or less concentration in 250ml flask the culture become yellow-green or yellow for 2 days, then some time it become green some time it remain yellow. I have tried by diluting media and changing pH and even BG11 (NaNO3 17.6 mM) but i did not getting constant blue-green culture from inoculation day.

You might have a phage issue (killing some of your Cyanobacteria) or they might lack some nutrients. Also, what is your light source and how strong is it?

As I have experience in working and maintenance of algal cultures in relation to one of the devastating tea pathogen, Cephaleuros spp. responsible for causing red rust in tea. I have used a number of media for the isolation and culture maintenance of the algal pathogen. I have also attached the publication, as it may help to standardise the algal culture media for your laboratory purpose.
