I was supposed to extrude ~100nm DPPC liposome via the Avanti extrusion kit after hydrating the liposome film.
However, it gave me bigger LUV during several trials (around 130-150nm).
Although it seemed there wasn't a noticeable aggregation, is there any common reason for making bigger-sized LUV than expected?

How many rounds of extrusion you perform?
Do you maintain temperatures above Tc?
How many times did you extrude the sample back and forth?
the larger size may result from only doing a couple extrusions.
i would recommend creating more samples with extruding back and forth different amounts.
Then use DLS to conclude the overall size of each sample and amount of extrusions.
This way you can find the optimal amount for you research.
Other reasons could be:

- old 100nm filters
