Any one can suggestthe time kill assay for herbal extract against bacteria?

I am trying to set protocol for Time kill assay but I get a variable results so give me a proper suggestion to set this protocol.

Can you describe your current protocol this might help to give some advice.

Do you get variable results with identical extracts or with differnt lots of extracts extraxted at different time points?.
