Hi everyone,
I have a blood sample that was RBC-lysed with ammonium chloride solution and frozen down in FBS with 10% DMSO in multiple vials. For the reason of the atypical composition of the WBC and the high cell number in each vial, I am thinking of separating the PBMC and the granulocytes before sorting for the cell type that I want.
Has anyone had any experience on using lymphoprep or ficoll on frozen WBC?
I know that the best way is to do it with fresh blood but I was given the sample in the way they are. There is also no way of gaining fresh blood. It would be really helpful if someone with the same experience can share with me.
