i have used crude extracts of plant lectins in PBS buffer for the hemagglutination activity. i have observed an increase in absorbance of supernatant (after agglutination and centrifuge) compared to suparnatant of control (only RBC). what does it indicates ?
Well, how "crude" were those extract? Any chance that you extracted something which absorb at the WL you are using?
Thank you Mr. Fabrizio, yes it could be because of that i have already used same amount of crude to check the absorbance and subtracted from the O.D but still it is much more higher ..
You mean the crude extract itself has lower absorbance of the supernatant after the hemagglutin experiment?
Some lectins next to hemagglutination activity may also induce hemolysis. In this case RBC contents (hemoglobin) would be released to the supernatant and would increase the absorbance.

Is there any small molecular weight Ligand which can bind with the protease enzymes (glycoproteins) without any adverse effect on Ligand-Ligate complex.
