I'm looking for counting platelets using flow cytometry (Vi-Cell from Beckman Coulter). Is there anybody who could help me with any advices concerning blood sample preparation, flow cytometer parameters
Surely the ViCell is a cell counter not a flow cytometer and is more often used to count cultured cells such as CHOs? It works by trypan blue exclusion for viability. I'm not sure it's usable for platelet counting. Given the relatively small size of platelets versus other cells, I would be surprised if it could differentiate between them and debris but I would be interested to seeif it can be done.
Coulter has a kit to perform cell count with known beads coun. In addition one can use CD41a, CD42 to evaluate platelets. I am not sure if your ViCell is a flow cytometer or not.
ViCell's are not flow cytometers. At our institute we do not use the Vicell for platelet counting as mentioned by others this is a tool often used for cell viability and is not very suitable to platelet enumeration due to their small size. An impedance counter can work well (often found in a hospital core lab). For truly enumerating platelets by flow cytometry it will depend on the flow cytometry machine you have available, some have the ability to provide counts/volume else this could be estimated by weighing tubes pre and post measurement as long as flow rates are steady. Consulting a local flow cytometry expert would be helpful to get you started.
Thank you very much for your kind answers. Saleh, my ViCell is not a flow cytometer but a cell counter as precised by Barry and Mark. I will follow your advices and I will use cytofluorimetry which is more suitable for platelets numeration.
