I need to assess fungal biomass (dry weight) as one of the parameters that would be studied in a large-scale experiment (150 bottles). I would use 25 ml broth/bottle and the experiment would be incubated in static conditions.
So I wonder if I can use 100 ml glass bottles instead of 100 or 250 ml Erlenmeyer flasks. I have some concerns about losing the surface-to-volume ratio of a culture that is facilitated by Erlenmeyer flasks to optimize oxygen uptake by the culture that might influence fungal growth!
The fact that you plan to incubate the containers in static conditions will have a bigger impact on the oxygen transfer into the cultures and the growth of the fungi than the shape of the container. Beyond that there is not enough information in the description you have provided to say whether or not bottles can be suitable for your experiment. In general, however, it can also be worth considering splitting your large experiment into smaller subsets of experiments - problems or errors are more likely to occur when trying to manage very large experiments.
