Anyone know of good antibodies that will mark B cells, dendritic cells or macrophages for immunofluorescence? Good staining in mouse brain would be optimal. Thanks!
No special antibody for immunofluorescence staining of brain cells. Only problem will be non specific staining of brain tissues. In order to minimize non specific staining of brain tissues, you had better perfuse mice heart by fixing medium to remove blood as much as possible from brain. B220 for B cells, CD11c for cDC, F4/80 for macrophages.
Thanks! We have perfused these animals with PFA and will be blocking with BSA to reduce nonspecific staining. I'll check out antibodies for those markers.
you need a live/dead cell stain, then CD45 is very helpful to get a positive signal for gating... then go on with cd19/B220, CD11b/CD11c and F4/80... good luck!
