I got essential oil using soxhlet extraction with pet benzene solvent. For Anti microbial study , my EO is not soluble with methanol, acetone and I can’t prepare stock solution using DMSO as well. But it is soluble in n Hexane,so can I perform the study using n hexane control
Yes you can use Hexane. Hence this is esential oil, it is bit difficult to dissolve in methanol, acetone and DMSO. But if this was a plant extract you can obtain the each extract dry powder bby evaporating each extract, then you can disssolve in DMSO.
Although, Hexane disolved EO extract can use for your microbial assay, sometimes it may give rise to false results. Therefore, if you can go with DMSO disolved solution it would be better.
However, you can go through the literatures and find out the morethings.

Yes you can use nHexane as a control for antimicrobial assay.
for conformation diluted 0.5McFarland bacterial broth culture transfer into control (nhexane) and check the turbidity after 24hr.
