If I want to analyse the plant extract by GC-MS, should I analyse all extracts differ by solvent (i.e. aqueous, methanol, acetone) or just the aqueous one.
I have been working with herbs for about a year now. GC I was using was equipped with HP-5MS 30 meter column and I was instructed never to use any polar solvent while doing my analysis because it can damage the column filling. I fixed the problem by doing an extraction with pentane and drying the extract with salts. I then evaporated the pentane. I took a small amount of non evaporated liquid and diluted it in pentane so I do not have a concentrated solution that wont give me clear peeks on the chromatogram. This method can be used if polarity of your solvent is much higher then polarity of the pentane so they form two layers necessary for an extraction (so water and methanol is ok, try it without your analit to see if two layers are forming).
