We extracted free amino acids (aa) from leaf tissue, using the 80-60-40% alcohol extraction method. Then quantified aa from the same samples by the Ninhidryn method and by GC-MS. The quantitation was completely different ( Coef Correlation 0.47) Has anyone done this comparison with the same results? Thanks

I did not use ninhydrin test for a long time, but as far as I remember it is not very precise, because you can't exclude a presence of free amines and small peptides in your sample. In GC-MS you can get relative abundances of individual AAs separately. But GC-MS also has its own limitations. So, basically, I would not be surprised in a low coherence between the techniques.
