Hello everybody,

Concerning the coupling reaction via amide bond formation between an carboxylic acid (CA) bearing metal complex and low molecular weight chitosan, which conditions would you prefer? As the complex itself is very good water soluble I was thinking of CA activation in water using EDC/NHS. I read a lot about buffer needed within this reaction due to pH dependency, but is this really necessary when I activate my CA and subsequently add my chitosan? And talking about adding, is it working when I add just the chitosan powder hoping that the product gets soluble? Or do I have to dissolve the polymer first in the usual 1% acetic acid?
