We have recently started culturing the alpha-TC1.6 cell line in our lab, however it took more than two weeks for the cells to grow to 50% confluence upon initiation, despite following what has been described in the ATCC protocol as well as the protocol described in paper published (e.g. correct media composition, handling procedure and culturing environment). We further waited for another 2 weeks for the cells to reach 70% confluence and split the cells in a ratio of 1:3, yet it took more than two weeks for the cells ready to be split again, and it has been the case in subsequent subcultures. There have been many reports showing that the cells are ready to split every week and could use up to late passage numbers (i.e. greater than 20). We are wondering if we have missed anything critical in the cell culturing protocols that led to the slow growth of these cells or if that is the property of these cells to be very slow growing?
