After performming the below protocol the percentage of recombinance I got is 60%. How can I extend the percentage of recombinance to 90?

How can I increase my recombinance

You will always loose some DN in a purification step. You should consider that you may underestimate your yield. You probably got rid of RNA small DNA fragments or nucleotides in your original sample.

Alternatively you may test another (e. g. spin column based) system.
