I am doing a validation test of an I-ELISA for B. canis and I experimented some problems with the ABTS, the negative controls are getting to high ODs. Have you experienced problems like this? How do you solve it? Do you have some advices on the basic use of ABTS?

Why don't you use TMB? There are very good ready-to-use substrates on the market, which should be essentially risk-free. If you still should encounter problems with the negative controls, you have an issue with other reagents, which might show some non-specific binding. This can occur if the concentration of your peroxidase conjugate is too high.
