I want to preserve my isolate of actinomycete for long term preservation, how feasible is this and after how long an incubation should it be preserved in glycerol?
In case of Actinomycetes, Mycelia glycerol stocks will lose viability in long term storage. It is recommended to store the strain as spores. I suggest the article, Shepherd MD, Kharel MK, Bosserman MA, Rohr J. Laboratory maintenance of Streptomyces species. Curr Protoc Microbiol. 2010;Chapter 10:Unit 10E.1. doi: 10.1002/9780471729259.mc10e01s18. for the same.

Manjula Shantaram Hi! The glycerol is used as a crioprotector and avoid the formation of crystal because it has a lower freezing point temperature. f you want to know more about the role in proteins you could check this article.
