Hi,

I am frequently working with human vitreous samples to perform western blotting. Most of the time, I prepare a fresh sample, heat and load them on the same day. By this way, I gets a regular thick fluffy band at the location of 60-70 kDa.

But, today, I loaded a vitreous sample that was prepared and heated 4-5 weeks ago. To my surprise, the blot comes out fine but devoid of the regular thick fluffy band at 60-70 kDa.

Is it bcoz of old stored loading of the sample or something else?

If someone can share & mention their experience to load a stored or previously heated samples for WB.
