I am facing an issue related to horizontal smearing on SDS gel. I have changed all the buffers and acrylamide with fresh ones, but I am still facing the issue. Below, the image is attached, where you can see the horizontal smear (thin line) appearing at the end of the gel. Other vertical smears in some wells are due to samples, but the horizontal one appears in every gel. Can you please provide a solution to solve this issue?
I usually use pre-made gels (supplied by Thermo). I also often get horizontal lines at the bottom of the gel. In my case it is yellow, but it does not prevent me from interpreting my results or taking the next step. It just makes me unhappy when preparing the graph. This may also be the case for you. Don't worry too much about the line. It will go away one day.
Young Eui Jeong: In my case, the interpretation of results is being disturbed because my protein is 15.2 kDa. So, the line is exactly where the probable band will appear. So it's an issue for me at least
