I am beginner for medical research and I need to make agarose gel for my lab member. Today my professor use that 0.8% agarose gel but I am not sure what samples he load and the samples mix with loading dye cannot pass through the gel and i am wondering that what cause it? So, I test with another 0.8% gel and i load only marker dye in one row and loading buffer in another row. I found that marker and also loading dye can pass through the gel. But he is angry that I make a wrong gel. My question is that can such a condition that sample cannot pass through the gel but control can pass through it. Thanks for Your Kindness. I am international student and I cannot get any support and kindness here.

Try to use a another electrophoresis device. That one might have the electrodes broken. Is the current passing throught?

if the loading dye moves in the right direction then the power supply and tank are working.
If this is human genomic dna that you are running then it is huge and will barely move into the gel and this is normal and as it should be..Only dna less than about 30000 bases and below will separate on an agarose gel and the best separations are with less than 5000 bases
Another possibility is that you forgot to add ethidium bromide then you will see the blue dye fine but you will not see the dna at all even if it runs perfectly
