In DCC coupling reaction of acid and alcohol, DCU is formed but many times DCU was not formed, the reaction did not give any product. Why?

I was doing a DCC coupling reaction of an acid and alcohol, and sometimes a product formed, but many times DCU has not formed even in addition of 1 equivalent of DMAP or HoBT. Why was this happening?

Can any one suggest any protocol for SEM sample prepration of solution of liposomes?

I have to take SEM images of a solution of liposomes, But i dont know how to process sample preparation. Can anyone suggest me reference paper? please help me to sort out this.
