I have to work on meiosis in the diploid yeast strain SK1. I have 1 gene which I want to tag with the AID cassette and study the effect of its depletion. I have amplified and purified the AID cassette with the homologous regions for my gene of interest (C-Terminal tagging). I have the diploid SK1 strain in my lab. However, now I am unsure on how to proceed. Initially I was thinking of doing AID separately in haploid W303 Mat-a and Mat-alpha and then mating them together. However, since SK1 is diploid how to proceed with AID approach in a diploid yeast strain? Will the AID cassette be integrated in both the alleles of my gene of interest? How do I confirm that both the alleles have been successfully tagged?
