My study is based on pretreatments and enzymatic saccharification to convert cellulose to glucose in lignocellulosic biomass. I tried several pretreatments which are liquid hot water, very dilute acid hydrolysis and mild alkaline pretreatments (for delignification) to enhance enzymatic saccharification. I can increase % cellulose content with these pretreatments in woody biomass. Also I am using commercial enzyme (cellulase) for enzymatic saccharification. However, I cannot enhance glucose concentration and I obtained that maximum 50% glucose conversion. How can I increase this conversion?

I suppose that you are already working at the optimum and stable conditions of pH, temperature and time. You could try to vary the ES ratio to be sure that the enzyme is saturated with the substrate and also vary the shaking rate of your reactor.
Also, this can be due to a feedback inhibition of enzyme by the products of the reaction. So you could try to make a separation when you reach 50% conversion.

The chemical compositions after pretreatment and enzyme type with enzyme dose would be important factor to convert cellulose to glucose. Furthermore, high solid loading tends to low conversion rate in short saccharification time. Please try to consider those criteria with Mr. Pierre Nobosse suggestion.

There are many things that could be happening, per example which concetration of cellulases are you using?. For woody biomass, I would suggest others pretreatments approaches such as steam explosion or dilute acid with higher severity.

Efficient saccharification of wood biomass cellulose may require more than just cellulases, but also other enzymes working synergistically to unravel the lignocellulolosic structure as the cellulose is intertwined and covered by other components such as hemicellulose and lignin which could hamper cellulase access to the cellulose thus the low cellulose yields.
